Effective hand washing is time consuming. To speed up this process, hand sanitizers that can be rubbed simply to the skin without the need of rinsing has been developed. They generally involve the use of alcohol as anti-microbial agent. However, there are several disadvantages with alcohol based hand sanitizers. Alcohol dries the skin . So, its repeated use can lead to skin damage. Application of certain material in cream formulation to skin would offer very promising vehicle for drug delivery for hand sanitation for those who has sensitivity to alcohol.Three new formulae were investigated and the most effective and safe cream formulation against bacteria, viruses and fungi was achieved. Rheological and pH measurements and organoleptic characterization of the cream were done. Formulation F1 showed non newtonian shear thinning flow behaviour and pH matches the pH of normal healthy skin. Its appearance was normal with non greasy feel or grittleness. These findings suggesting it as the formulation of choice.Cytotoxicity experiment was done to determine the safe dose of the 3 formulae of the cream. All were found to be safe.The cream have in vitro antimicrobial activity against Pseudomonas aeruginosa, Pseudomonas putida, Salmonella Typhimurium, E. coli, Staphylococcus aureus, Listeria monocytogenes and Candida albicans using disc diffusion method. On the other hand, the cream was tested in vivo as a hand sanitizer for 15 volunteers with different ages and genders. The results showed that, the removal percent of total bacterial counts ranged between 91 and 100%. Furthermore, the removal percent of total fungi ranged between 95.38 and 100%.
The effect of cream on the infectivity of food-borne viral surrogates, Phix-174 (ssDNA) bacteriophage and simian rotavirus sa-11 (dsRNA) was evaluated. The viral titer after each treatment was evaluated by standard plaque and TCID50 assays. The mean log10 reduction values for Phix-174 and rotavirus were 0.02 and 0.1, respectively after 30 min. In conclusion, this cream can be used as a safe, effective and cheap antiseptic for hands and can replace alcohol in the market generously
